FLIM Guided Nanomedicine: PLGA Nanoparticles for Controlled Baricitinib Delivery

Introduction:

Diabetes is characterized by progressive β-cell dysfunction and impaired insulin secretion1. Chronic inflammation and JAK–STAT signaling drive β-cell stress, making immunomodulation a potential strategy. Baricitinib (Bar), a selective JAK1/2 inhibitor, may reduce inflammation-driven damage2. Encapsulation in PLGA nanoparticles improves delivery and release control. Fluorescence Lifetime Imaging Microscopy (FLIM) enables non-invasive characterization of encapsulated drugs, distinguishing free and loaded forms and providing insight into supramolecular organization without labeling3.
