Given the significant role and growing popularity of lipid-based nanocarriers, especially since the COVID-19 pandemic, we propose an analytical method for the straightforward and reliable physicochemical characterisation of PEGylated lipid-based nanoparticles. This approach integrates size separation using Asymmetric Flow Field-Flow Fractionation (AF4) with composition analysis via High Performance Liquid Chromatography coupled to a Charged Aerosol Detector chromatography (HPLC-CAD). The rationale behind this strategy is to exploit AF4 for isolating liposomes from unincorporated molecules, thereby enabling reliable quality control regarding their composition.
