Introduction: Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal cancer that is often diagnosed after metastasis, limiting surgery and resulting in poor survival. Although gemcitabine is a standard therapy, its efficacy remains limited, and pressurized intraperitoneal chemotherapy also has practical and distributional drawbacks. Rapamycin may enhance anticancer activity, but its poor solubility restricts application. Therefore, this study aimed to develop an injectable thermosensitive PLGA-PEG-PLGA gel co-loaded with gemcitabine and rapamycin for localized intraperitoneal treatment of metastatic pancreatic cancer.
