Glioblastoma (GBM) accounts for nearly half of all gliomas and remains associated with poor prognosis due to limited brain penetration and systemic toxicity of temozolomide (TMZ). Targeted nanocarrier systems offer a promising strategy to overcome these limitations. This study aimed to develop biotinylated PAMAM (G3.0, NH2) dendrimers conjugated and encapsulated with TMZ to enhance cellular uptake, controlled drug release, and brain biodistribution for improved GBM therapy. This might potentially lower the high dosage needs and related adverse effects of traditional TMZ therapy.
