Brain cancer treatment remains challenging due to the limited ability of most chemotherapeutic agents to cross the BBB. Everolimus, a potent mTOR inhibitor, has poor solubility and inadequate CNS penetration, which restricts its clinical efficacy. Nanoparticle-based delivery systems utilizing biodegradable polymers offer a promising approach to overcoming these limitations. This study aims to focus on the development of hyaluronic acid-conjugated, chitosan-coated, Everolimus-loaded polycaprolactone nanoparticles (HA@CS@EVR-PCL NPs) for targeted delivery (CD44 Receptors), pH-responsive release, and therapeutic performance.
