Glioblastoma (GB) is the most lethal brain tumour, where the blood-brain barrier (BBB) and an immunosuppressive environment render standard therapies ineffective. While immunotherapies are promising, systemic delivery lacks specificity, leading to off-target toxicity and suboptimal therapeutic concentrations at the tumour site. To address this, we developed an Aptamer-Targeted Lipid Nanoparticle (Apt-LNP) platform that utilises synthetic single-stranded oligonucleotides. These LNPs recognise specific GB biomarkers to facilitate receptor-mediated transcytosis across the BBB, enabling precise drug delivery and enhanced therapeutic efficacy.
