Introduction: Efficient analytical characterization of adeno‑associated viruses (AAVs) is critical for process development and quality control but remains time‑consuming with established methods. Techniques such as analytical ultracentrifugation (AUC) require several hours, while liquid chromatographic approaches including anion exchange chromatography (AEX) size exclusion chromatography coupled to multi‑angle light scattering (SEC–MALS) typically require ≥30 minutes per analysis and additional time to reach stable operating conditions. Here, we report a high‑throughput SEC–MALS workflow enabling the simultaneous determination of AAV fill state and titer within 8 minutes per sample.
