mRNA holds huge promise for future medicine and lipid nanoparticles (LNP) is currently the most advanced delivery vehicle for RNA drug. However, comparing to mRNA vaccines, delivering mRNA for therapeutic purpose requires higher translational potency (to produce more therapeutic proteins) and extend circulation half life (to achieve extrahepatic delivery). This report will discuss key structural features of LNPs and their indication in both potency and pharmacokinetics.
