The subcutaneous route has gained increasing interest for extended drug delivery in chronic therapies, as it allows prolonged exposure, reduced dosing frequency, fewer side effects, and improved patient compliance. In situ–forming thermosensitive hydrogels, particularly when combined with versatile nanocarriers such as DELOS nanovesicles (DELOS-NV), capable of encapsulating therapeutics from small molecules to biologics, offer a promising strategy to create extended-release depots, enabling self-administration, avoiding first-pass metabolism, and overcoming poor oral bioavailability.
