Intranasal MK-801 delivery and LC-MS/MS quantification in plasma and brain following concussion in rats

Introduction: MK-801 is a non-competitive NMDA-glutamatergic receptor antagonist used in rodent models of traumatic brain injury (TBI) (1). Alterations in amino-acid concentrations are major contributors to the persistent neurological and behavioral effects induced by concussions and mild traumatic brain injuries TBIs (2, 3). This study aimed to establish an integrated formulation and bioanalytical platform to support the investigation of MK-801 as a treatment agent against concussion.

to describe formulation strategies for intranasal delivery of poorly soluble compounds in rodent models.
to evaluate LC-MS/MS approaches for quantifying drugs and endogenous amino acids in plasma and brain tissue.
