Gut probiotics have been reported to ameliorate gut disorders. However, the use of live bacteria raises stability and safety concerns. Here, we introduce a cell-free, gut-targeted strategy based on probiotic-derived outer membrane vesicles (OMVs), which are termed as PO. Briefly, OMVs are biomineralized with tannic acid (POT) and encapsulated within pH-responsive alginate microbeads, forming an engineered platform (POTA) that amplifies their anti-inflammatory and antioxidant activities while ensuring intestinal release. We demonstrate that POTA significantly alleviates intestinal barrier damage and inflammation in inflammatory bowel disease (IBD).
